Bioscreen H2O2 titration

H2O2 titration in all Met15+/- strain.

Day 1, Feb 14, 2011 Monday, Grow all strains in 5ml YPD @30C shaker, overnight.

Day 2, Feb 15, 2011 Tuesday, Bioscreen titration

Measure OD600nm

1:1000 Dilution in YPD,  all strains to OD<0.03.

198ul yeast mix

2ul 100x H2O2 solun:       30%    20%    10%     5%      2.5%     1%      0.5%    0.25%   0%

Final H2O2 solutions of 0.3%, 0.2%,  0.1%,  0.05%, 0.025%, 0.01%, 0.005%, 0.0025, 0% ,

For standard curves: use 3fold serial dilution of YPS128 and M2-8. (200+400 dilutions)

For halo assay, add 5ul 30% and 15% H2O2 to the center of each YPD plate. Measure inner and outer diameters.
